Introduction There is always more than one way to do something and education is no exception. Educators have different philosophies and classroom approaches depending on their subject and the type of students that they have. This semester, I had the opportunity to apply information from a class to real world situations. Through my field experience at the Whelchel Family Life Center, I was able to see the challenges associated with teaching different social classes, different demographics, and different learning styles. I was able to learn how to navigate through common challenges from observing my mentoring instructors, which in turn will help me to develop my own teaching style and better prepare me to be a future educator. Whelchel Family Life Center The Whelchel Family Life Center is a non-profit center whose goal is â€Å"to provide services to the community by enriching the spiritual, educational, physical, emotional, and social needs of youth, elderly, and families of the local and surrounding communities† (W.L.). This center is used to promote health for all members of the community in need of support mentally or physically. I worked closely first through eighth graders. There was a variety of both male and female students. The demographics was mostly African American students and mentors, but some Caucasian students and volunteers. The once successful UK Company that only specialized in home decor is now trying to tap in the garment design, expend every line of selection, and rapidly grow internationally, especially in North America where the company does not have a good understanding of the market place. The company needs to consolidate the past and existing customers: middle to older aged women, and will benefit from the aging world population. The Winning Formula for Writing Success The Winning Formula for Writing Success The Winning Formula for Writing Success By Mark Nichol When I wrote the heading for today’s post, I thought to myself, â€Å"I should be making infomercials and workshop presentations, offering my ‘secret’ for a thousand dollars.† A thousand dollars a head for even a few dozen participants? That’s what I call successful writing: With one phrase and a few platitudes, I could take a couple of years off from work. Nah. I’ll give it to you free of charge: Quality requires quantity. Yes? And? That’s it. Quality requires quantity. Oh, all right. I’ll expound. That’s a layered statement, one that’s as deep as you want to dive. But on its most basic level, it means that an output of high quality must be preceded by an input of high quantity. In other words, a return of quality takes an investment of quantity. The new publishing model is that, thanks to the Internet, everyone’s a writer. That’s the good news. But it’s also the bad news, because it means that because many writers in this suddenly expanded universe are not highly qualified, the universe is degraded. There have always been less-than-stellar writers, but it was more difficult for them to publish their work and sustain success. Now, however, nonprofessional writers can be forgiven for believing that because it’s easy to type, it’s easy to write. And the remaining exemplars of great writing are lost in the leveling of the signal-to-noise ratio if they are sought out at all anymore. The brave new world of formal publishing is also degraded, in this case by a business model that no longer values quality because remember, quality requires quantity (and quantity, of course, requires financial investment). So now, I can find six typographical errors stuffed into a twenty-word caption in the website for a major metropolitan newspaper (since corrected because, hey, it’s the instant Internet, and we can always fix it later!), and I can find my enjoyment of a newly published book compromised by shoddy editing (improvement of which must wait for the second edition, if there is one, and if there is the wherewithal to improve it but that’s too late for me). That’s why I may come across as a dinosaur about these things, because I believe that if something is worth doing, it is worth doing well. And because I believe that, that’s why I’m proud to remain part of the old-school old guard, editing book manuscripts for publishers willing to spend time, effort, and especially money to ensure that their products reflect their high standards. Quality requires quantity. Oh, quality is sometimes accidentally produced with a minimum of quantity, but standards cannot rely on serendipity. The work ethic is called that for a reason: Good isn’t easy. It takes effort. Quality requires quantity. There’s at least one other layer to the formula. Last week, I wrote a post about another formula, what I call a writing-competence matrix. Rather than explain it here, I invite you to read the post, if you haven’t already, or to review it, if you have. Go ahead. I’ll wait. What does it take to score high on this matrix? Don’t expect to ever hit â€Å"Expert† on all three counts; many successful writers may excel in only one category. But to rate highly in even one area takes time. Remember the 10,000-Hour Rule? (That’s all right. I’ll still be here when you get back.) If you want to be a great writer, be content at first with endeavoring to be a good writer. Great can wait. But to become a good writer, you must invest quantity in your quest for quality quantity of time and effort. And you must be willing to labor not only longitudinally, putting in years of skill development, but also latitudinally, massaging, refining, and polishing each piece of writing along the way. Minireview of A Study Essay Synaptic transmission in vertebrate neuromuscular junctions forms the basis of this study. The communication between neurons in the nervous system occurs largely due to neurotransmitter release at the synapses. Messages on the various significant nervous system functions are coordinated through the synaptic junctions and the release of neurotransmitters. Ryanodine receptors are found in the somata of the Purkinje cells, basket cells and pre-synaptic terminals of specific synapses and terminals of basket cells. Calcium in the extracellular fluid triggers the neurotransmitter release. Now it is understood that pre-synaptic stores could participate in this same function. Ryanodine-sensitive stores of calcium also play a role. The quantum is the amount of spontaneous signals occurring in the absence of pre-synaptic action potentials and is equivalent to the release of one neurotransmitter vesicle (Katz, 1969). The action potentials are called miniature currents. For central synapses in the brain, large miniature currents are believed to arise from the release of many neurotransmitter or presynaptic vesicles and in the range of several quanta (Bekkers, 1994). Yoshida’s study (1994) revealed that these multivesicular miniature events could actually be tetrodotoxin-resistant action potentials in the pre-synaptic terminals. Other researchers have tried to explain the phenomenon from another angle, using the presence of intracellular calcium stores in the pre-synaptic terminals. Nakanishi et al localized inositol triphosphate receptors in the neural tissue of the developing and adult mouse brain (1991). These were immunolocalised in the pre-synaptic terminals of the deep cerebellar nuclei and the retina of the eyes. Narita’s studies (1998, 2000) revealed the action of ryanodine-sensitive calcium stores at the frog neuromuscular junctions. It was discovered that agents which influence the ryanodine-sensitive Calcium stores also increased the intracellular Calcium in the pre-synaptic cells and regulated acetyl choline release during high frequency stimulation. Mothet et al (1998) studied the action potentials at the pre-synaptic terminals of the buccal ganglia in Aplysia. They indicated that ryanodine inhibited while the pre-synaptic injection of Cyclic ADP Ribose augmented the action potential evoked release of acetyl choline at synapses. Studies also showed that caffeine with or without ryanodine modifies Calcium stores at the pre-synaptic terminals in autonomic ganglia (Peng, 1996; Smith et al, 1996) and in photoreceptors (Krizaj, 1999). Studies on hippocampal pyramidal cells have shown that Caffeine or thapsigargin influences the frequency of miniature IPSCs. Making an assumption, from prior studies described above, that spontaneous Calcium release from pre-synaptic Calcium stores may provide the synchronisation mechanism that causes multivesicular miniature IPSCs and the fact that such a hypothesis has not been tested systematically previously, the authors have taken up this topic for their study on cerebellar interneuron Purkinje cells. This Study   Methods Experiments were conducted on the sagittal cerebellar slices of decapitated rats aged 10-14 days of age. During the experimental recordings, the slices were perfused in saline containing prescribed concentrations of NaCl, KCl, Na H2PO4, NaHCO3, CaCl2, MgCl2 and glucose with 95:5 mixture of oxygen and carbon dioxide. Experiments were done at room temperature. For tight-seal whole-cell recordings, pipettes filled with a solution of appropriate concentrations of CsCl, MgCl2, HEPESCs, BAPTA-Cs (Molecular Probes, Eugene, Oregon), CaCl2, Na-GTP and Na-ATP and of pH 7.3 were used. Capacitance cancellation and series resistance compensation had been done. Kynurenic acid had been added to the extracellular solution to block the inotropic gluatamate receptors. TTX was present in the solution for all recordings. The calcium free solutions were prepared by leaving out Calcium and adding EGTA Na. Membrane potential was maintained at -60mV and the current was filtered at 1.5-2 kHz. Sampling was done continuously with brief interruptions. Detection and analysis were done using the IGOR-Pro programming environment. In experiments needing a Calcium channel blocker, cytochrome was added to the external solution. The toxin was prepared while the ryanodine was purchased. Testing the Calcium The Calcium in the basket cells were tested using the Two-photon laser scanning Fluorescence microscopy. For studying the action potential-evoked calcium increases, bicuculline was added to the external solution and the calcium sensitive probe Oregon Green was put into the pipettes. Scans were done and pulses were applied at the end of each 8th scan. This was repeated every minute in external solution which contained saline in order to get a baseline. The external solution was then changed to the solution containing ryanodine and recording proceeded for another 15 minutes. Another set of recordings were done with external solution not having calcium but having EGTA Na. The internal solution also had EGTA and Cs instead of K as the main cation. The responses in Spontaneous Calcium transients also were recorded using molecular probes in the external solution. A pseudo line scan was also done. For immunocytochemistry, a rabbit polyclonal anti-serum was raised to the 16 amino-acids found in all mammals. A C terminal cysteine enabled conjugation to haemocyanin. The conjugated peptide was used to immunize rabbits. Then the ELISA confirmed the specificity. The sarcoplasmic and endoplasmic reticulum microsomes derived from the skeletal muscle, cardiac tissue, whole brain and cerebellum were used for immunoblot analysis. Effects of external Calcium concentration. Large amplitude miniature IPSCs in cerebellar Purkinje cells were found sensitive to extracellular Calcium. With calcium free solution, the mIPSC frequency fell suddenly to half the control level. Continued exposure to the low level of external calcium caused the frequency of mIPSC to continue declining but at a slower rate. On washing after this, the frequency recovered and reached its initial level. The amplitude of the mIPSCs on the other hand showed a steady decline all through and no recovery on washing. With high levels of Calcium, the frequency of the mIPSCs increased rapidly and significantly. The change in amplitude varied from no response to a minimal increase. The inference was that Calcium strongly influenced mIPSCs in the Purkinje cells though frequency and amplitude were differently affected. The rapid change in frequency was interpreted as the reaction of intracellular Calcium to external Calcium changes. The slow change in amplitude was considered due to the extracellular influence on the pre-synaptic stores. The prolonged extracellular calcium removal could have caused selective elimination of large amplitude miniature IPSCs. Repeating with a calcium-free solution, many large amplitude miniature IPSCs were seen again. Then there was a sudden drop and then the amplitudes reduced to become concurrent with the control and the IPSCs were also less. The reduction seen when external Calcium was removed was not due to post-synaptic modifications. On returning to the calcium-rich solution, a slight recovery of both amplitude and frequency occurred. Paired Student’s t-tests indicate significant changes in mean amplitude and frequency between mIPSCs recorded during a 3-min control period and after 15–18 min in Calcium-free external solution. 6 sham experiments were also conducted by keeping the slices in Calcium containing external solution all throughout and these showed no obvious change. The time course of decay of the IPSCs was slower in calcium-free external solution when compared to depolarization-induced calcium transients. Effects of elevated intracellular Calcium Elevated intracellular Calcium in the Purkinje cells caused a speedy frequency reduction and a slow increase in amplitude. This sudden fall could not be explained by the intracellular calcium as BAPTA buffered the Calcium in the cells and could not have caused the IPSCs. Effect of the axons in large amplitude miniature IPSCs The immediate slow changes in the amplitude of IPSCs on withdrawal of external calcium for 3 minutes could not be attributed to the delayed removal of external calcium from pre-synaptic release sites. The effect was also not due to local Calcium influx caused by the TTX insensitive axonal depolarization. For the latter test, external Cd,   a non-selective channel blocker, was used. It reduced the action potential-evoked pre-synaptic Calcium transients seen in the axons and pre-synaptic terminals of cerebellar interneurons. The slow changes in amplitude were therefore not connected to rundown or altered post-synaptic receptors or delayed extracellular calcium removal. The only remaining explanation was that multivesicular release under the pre-synaptic calcium stores could have caused the changes in amplitude. The lack of recovery after external Calcium restoration could be due to the slow store refilling of intracellular Calcium when action potentials and subsequent calcium influx are blocked. Rise Time Kinetics The multi-vesicular release also could not completely explain the slow changes in amplitude. Rise time of IPSCs as a function of amplitude was studied after extended external calcium removal. In 6 of the 8 cells tested, the rise time was heterogenous in nature where two subpopulations were concerned. Slower decay kinetics was also noted. A faster rise time was seen in the proximal dendrites and soma. Faster IPSCs were more sensitive to external calcium removal than slow ones and these IPSCs arose at somatic synapses. The origin being multivesicular, synchronisation time must have been in the range of submillisecond. Some IPSCs had slower rise time and decays and were less sensitive to external calcium removal. These IPSCs could be arising from dendrites and could be having synchronisation of 1-4ms. Ryanodine receptors The two photon laser illumination was used to focus on the ryanodine receptors stained with a high-affinity Calcium-sensitive dye, Oregon Green BAPTA-1. Transient rises of Calcium in the stained ryanodine receptors in response to short trains of action potentials were measured. The fluorescence rises were noted. Ryanodine-sensitive Calcium stores are associated with the large amplitude mIPSCs. In the experiment Ryanodine in large concentrations of 100 ÃŽ ¼M blocked the receptors. The responses to muscimol were not affected by ryanodine and the researchers assumed that ryanodine had no post synaptic effect. Axonal spots with calcium stores were identified in pre-synaptic terminals by recording responses to short action potentials. The intracellular Calcium fluctuations in response to differing concentrations of external calcium to which small concentrations of ryanodine were added was checked. Repeated scanning in the presence of TTX was done. Spontaneous calcium transients were noted before and after addition of ryanodine. Bright spots of fluorescence were noted at the pre-synaptic terminals. This signified the presence and increase of Calcium at the pre-synaptic terminals. Experiments were then done with small concentrations of Ryanodine 10 ÃŽ ¼M. Large amplitude mIPSCs were recorded. High frequency bursts and amplitudes of the responses were greatly increased. The bursts could reflect the response at multivesicular and monovesicular sites. The histogram comparing the amplitudes in the control and after ryanodine was added showed a significant difference. In our class, Survey of American Literature, we have read and analyzed the classic American works that have influenced and in some cases ingrained themselves into our country’s identity and belief system. Among those studied has been Benjamin Franklin’s Autobiography, Ralph Waldo Emerson’s essays, The American Scholar and Self -Reliance, and Frederick Douglass’ Narrative. Each of these covers a vast amount of subjects that could be discussed, but one that proves most compelling is the topic of education. These three men, who come from different life circumstances and have varied philosophies of life, each came to define the uses of education in their own ways. However, they do all seem to express the notion that education can be used to empower one’s self. In Benjamin Franklin’s Autobiography, comes through to me as a perfect example of the American dream, that being the idea that no matter where you come from, you can achieve almost anything with hard work and dedication. Coming from a lower-middle class Boston family to being one of the most famous Americans of all time is the classic rags to riches story that he himself lived out. The way he achieved this was by being self-educated. When looking at Franklin’s life, it could be said that he is a Renaissance man because he did so many things; he was a printer, a writer, an inventor, a scientist, and a statesman, and the only thing he had a formal education of was of printing. This American dream that is still deeply ingrained in our society today though, always seemed to be a bit of a myth to me. I don’t think it is right to say that if you are willing to work hard enough, you will always achieve success. Things are not that simple. Of course hard work can get you places, but some people work very hard their entire lives and still don’t achieve success due to unfair circumstances. I would say that success is achieved not only by hard work, but also by the right circumstances and a bit of luck. It seems to me that Franklin did not view education as what is important in life. What is important to him is what you accomplish in life. He talks about success and how to achieve it, and education is only part of that. â€Å"I grew convinc’d that Truth, Sincerity and Integrity in Dealings between Man and Man, were of the utmost Importance to the Felicity of Life†¦Ã¢â‚¬  pg. 268, Benjamin Franklin, The Autobiography (Part 1), The Norton Anthology of American Literature. Not that education could be seen as unimportant to him. He did create the first ever circulating library, which is a way in which others can become self-educated. He almost certainly felt that education was of great importance. It is just that I think he viewed education as a necessary means to get to the greater goal of being a successful person. Education was a way to prepare oneself for success. Ralph Waldo Emerson was a Transcendentalist, and therefore had a somewhat different take on education. He believed that all people are inherently divine. According to him, we all know truth in hearts but we often don’t recognize it or express it. I think he wants people to listen more closely to themselves for answers to their questions. Education can be used to inspire us and help us realize our truths. In The American Scholar, Emerson says the true sources of education are nature, the mind of the past (most influential being books), and the action taken of the heroic mind. In nature he believes we see a visible representation of what is in your self, and that is ultimately divine. And because true wisdom lies within, books can help bring it out. What is most interesting to me is that of taking action with the knowledge you have. I think here he relates to Franklin in the thought that an education is no good if you don’t go and use it. His definition of action: â€Å"The preamble of thought, the transition through which it passes form the unconscious to the conscious, is action. Only so much I know as I have lived†¦Ã¢â‚¬  pg. 519, Ralph Waldo Emerson, The American Scholar, The Norton Anthology of American Literature. Without action, a person would never experience the life issues that they studied. That would make it impossible to fully understand or form an  opinion over almost anything. Gaining knowledge will only be important if it is applied through action, and validated by action. Emerson sees education as way to inspire your own mind, and in turn, help you on a road of self-discovery in your own wisdom. Frederick Douglass comes from the most difficult circumstances (to put it lightly), being that he was enslaved and a black American in the 19th century. His autobiography is a story of self-discovery; he details how his growing awareness of his condition, through his self-acquired education, propelled him to re-imagine himself. He had to learn to think of himself as a human being rather than as a beast of burden, as a free man rather than as a slave. This required him to educate himself at the risk of brutal punishment and then to take the even greater risk of an escape attempt. After one failed try that could easily have cost him his life, he succeeded the second time. Douglass’ story is one of self-reliance. To him, education was necessary to his survival because it offered him a first step to get out of a life of slavery. â€Å"I understood the pathway from slavery to freedom†¦Though conscious of the difficulty of learning without a teacher, I set out with high hope, and a fixed purpose, at whatever cost of trouble, to learn how to read†¦Ã¢â‚¬  pg. 946, Frederick Douglass, Narrative, The Norton Anthology of American Literature. Each of these men, Franklin, Emerson, and Douglass, viewed the need for education for seemingly different purposes. Franklin, who focused on how to be a successful person understood that education, was an important step in preparing oneself for future accomplishments. Emerson felt we all had the answers within, but saw education as a way to prompt our understanding of those inner truths and convince us to express them. Douglass used education for survival and a small ray of hope that he would not remain a slave forever. But what is true of all these men is that education was used to better themselves, they’re understanding of the world around them, and to then educate others through their literary works. To educate the self is to empower the self.
